Liquid phase or vapour phase?

Choosing the right storage method for the samples—not simply following a general preference

Liquid-phase freezers immerse samples in liquid nitrogen at close to −196 °C. Vapour-phase freezers keep the samples above the liquid, within the cold nitrogen vapour. Both can provide long-term cryogenic storage, but they manage temperature, contamination risk and access differently.

Why vapour phase is often selected

Keeping samples out of the liquid removes direct contact between the container and the storage medium. This reduces the risk of liquid entering a damaged or poorly sealed vial and removes one route by which contamination could pass between compromised containers.

It does not make the freezer sterile, and it does not prove that every sample position is cold enough. Vapour-phase equipment has a temperature gradient, so the warmest part of the usable storage zone and its behaviour during normal access need to be understood.

When liquid phase may still be appropriate

Immersion provides a very low, comparatively uniform temperature and substantial thermal reserve. It may remain appropriate where the approved storage process requires liquid-phase conditions and the sample containers, overwraps, retrieval method and contamination controls are suitable for immersion.

The questions that decide it

· What temperature range has been approved for the samples?

· Is direct contact with liquid nitrogen permitted?

· Are the vials, bags or straws and their closures suitable for the chosen method?

· How frequently will the freezer be opened and how long will retrieval take?

· Can the equipment demonstrate the required temperature across every usable position?

· What qualification, monitoring, alarm and backup evidence is required?

The freezer label is not the decision. The selected equipment must deliver the sample owner’s approved conditions in everyday use, with practical capacity, monitoring and resilience taken into account.
